Dose Dependent Morphological Effects of Alpha Lipoic Acid on Vasospastic Femoral Artery in Rats

Öz:
AMAÇ: Alfa lipoik asidin (α-LA) doza bağımlı etkileri bir sıçan vazospazm modelinde morfoloj ik olarak araştırılmıştır.yÖntem ve GereÇler: 32 sıçan dört gruba bölünmüştür: grup I = kontrol; grup II = vazospazm; grup III = vazospazm + düşük doz (20 mg/ kg) intraperitoneal α-LA uygulaması ve grup IV = vazospazm + yüksek doz (100 mg/kg) intraperitoneal α-LA uygulaması. Her grup için ışıkmikroskopu altında histoloj ik ve morfometrik incelemeler yapılmıştır.BULGUlAR: Ana vasküler duvar kalınlığı grup II ve IIIte grup I ile karşılaştırıldığında önemli bir artış göstermiştir (p<0,05). Grup II ve IVünistatistiksel olarak karşılaştırılması vasküler duvar kalınlığı açısından grup IVte önemli bir azalma ve vasküler lümen açısından yine grup IVteönemli bir artış göstermiştir (p<0,05). SONUÇ: α-LAnın yüksek doz tedavi grubunda duvar kalınlığını azaltıp lümen yüzey alanını arttırarak vazospazm etkilerini azalttığı gösterilmiştir.Mevcut çalışma yeterli dozda α-LAnın deneysel bir vazospazm modelinde potansiyel bir terapötik ajan olduğunu düşündürmektedir.

Öz:
AIM: The dose dependent effects of alpha lipoic acid (&#945;-LA) were investigated morphologically on rat vasospasm model.mAterIAl and methOds: 32 rats were divided into four groups: group I=control; group II=vasospasm; group III=vasospasm +low dose (20mg/kg) intraperitoneal &#945;-LA administered; and group IV=vasospasm +high dose (100 mg/kg) intraperitoneal &#945;-LA administered. Histologicaland morphometric examinations were carried out for each groups under light microscope.results: The mean vascular wall thickness displayed significant increase in group II and III compared with group I (p<.05). Statisticalcomparison of group II and IV, regarding vascular wall thickness showed a significant decrease in group IV, and regarding vascular lumen areashowed a significant increase in group IV (p<.05). COnClusIOn: It is demonstrated &#945;-LA reduces the effects of vasospasm in high dose treatment group by decreasing the wall thickness andincreasing the lumen surface area. The present study suggests that adequate dose of &#945;-LA is a potential therapeutic agent in experimentalvasospasm model.
