Maternal thyrotrophin-releasing hormone (TRH) administration accelerates maturation in fetal rat lung; A morphologic study by electron microscopy

Öz:
Prematür doğan fetuslar için bugün en önemli tehlike respiratuar distress sendromu gelişimidir. Bu çalışmada antenatal dönemde profilaksi amaçlı uygulanan TRH'nın fetal rat akciğerindeki maturasyon üzerine olan etkisini morfolojik olarak araştırdık ve kontrol grubu ile karşılaştırdık. Sekiz gebe rat İki gruba bölünerek gebeliklerinin 13 ile 18. günleri arasında birinci gruba TRH 20 mg/kg/gün, kontrol grubuna ise serum fizyolojik intramüsküler olarak enjekte edildi. Her iki grup 19. günde sakrifiye edilerek yirmişer embriyo toplandı. Işık ve elektron mikroskopu kullanılarak fetal akciğerlerin histolojik analizleri yapıldığında her iki grup arasında belirgin yapısal farklılıklar saptandı. TRH grubunda daha kalın bir mezenkim, daha ince bir septa, daha dilate olmuş alveolar odacıkları ve diferansiye tipte alveolar hücreler saptanırken konrol grubunda tersi gözlendi. Sonuç olarak TRH injekte edilen rat embriyolarının akciğerlerinde kontrol grubuna göre hem ultrastrüktrel farklılığın olduğu hem de surfaktan sekresyonunun oluştuğu saptandı.

Maternal thyrotropin salgılatıcı hormon (TRH) uygulaması fetal ratlarda akciğer maturasyonunu hızlandırmaktadir

Öz:
OBJECTIVE: Premature fetuses are at increased risk of neonatal respiratory distress syndrome. The effects of the antenatally-administered TRH for profilacsis on fetal rat lung maturation were investigated morphologically and compared with the control group. STUDY DESIGN: Eight pregnant rats, which divided as two groups that TRH 20 (ng/kg/day) injected and saline injected as a control during between 14th and 18th day of gestation, were used. Twenty fetuses from each group was obtained on day 19 of their gestation by sacrifiying the rats. Histological analyses of the fetal lungs were performed with both light and electron microscopy. RESULTS: Obvious structural differences were observed on TRH group when compared with the control group. Thinner mesenchyme, thicker septa, less dilated alveoli and undifferentiated-type alveolar cells were found in lungs of control group whereas the opposite is true for TRH-injected rats' fetuses. CONCLUSION: Our results demonstrated that when the embryo injected with TRH, both progressive fetal rat lung maturation with ultrastructural differences and surfactant secretion was detected.
