Genetik bağlantı analizi ve uygulama alanları

Öz:
Sirke sinekleri (Drosophilia melanogaster) üzerinde yapılan çalışmalarda, bazı özelliklerinin kalıtımının Mendel Kanunlarına uymadığı ve genlerin kromozomlar üzerinde bileşik olabileceği belirlenmiştir. Dolayısıyla, genetik linkage (bağlantı) kavramı ve ilk genetik bağlantı haritası başarıyla ilk kez ortaya konulmuştur. Gen haritalaması, genlerin ve markörlerin kromozomlar üzerinde bulunduğu yerlerin ve birbirlerine olan uzaklıkların tespit edilmesidir. Krossing-over olayından yararlanarak, bileşik genlerin dizilişi ve aralarındaki uzaklıkların bulunması sonucu genetik bağlantı haritaları oluşturulmaktadır. Genetik bağlantı analizi farklı uygulama alanlarına sahiptir. Bağlantı analizleri gen haritalarının oluşturulması ve türler arası karşılaştırmalı genom (comparative genomics) çalışmalarında kullanılmaktadır. Özellikle, ekonomik öneme sahip özelliklerin ve hastalıklara sebep olan gen bölgelerinin tespiti amacıyla genom tarama ve kantitatif özellik lokusları (QTL) haritalama çalışmaları ile markör destekli seleksiyon (MAS) uygulamalarının temelini oluşturmaktadır.

Analysis and applications of genetic linkage

Öz:
Research conducted on fruit flies (Drosophilia melanogaster) has indicated that inheritance of some characters did not follow Mendelian Laws; and genes might be linked to each other on chromosomes. Therefore, genetic linkage as a concept and the first linkage map were reported for the first time. Gene mapping terminology refers to the indication of positions of genes and markers; and the distances between them on chromosomes. Genetic linkage maps are formed by utilizing crossing-over events, following the determination of orders of the linked-genes and the distances between them. Analysis of genetic linkage provides various application possibilities. Linkage analyses are used for linkage mapping and comparative genomic analyses between species. In particular, they form the basis of the studies on genome scanning and quantitative trait loci (QTL) mapping aiming to quantify the traits that are economically important and the gene locations that cause diseases; as well as the applications of marker assisted selection (MAS).
