Epstein-Barr Virüsü ve Gastrik Kanserde DNA Metilasyonu

Öz:
Gastrik kanser (GK) tüm dünyada görülme sıklığı yönünden dördüncü ve kanserle ilişkili ölümlerde ikinci sırayı alan bir malignitedir. GK etyolojisinde Helikobakter pilori (H. pilori) ve Epstein-Barr virüs (EBV) gibi enfeksiyon ajanlar önemli rol oynamaktadır. EBV Herpes virüs ailesinden, yaklaşık 184 kb boyutunda, çift-dallı DNA virüsüdür ve insan neoplastik hücrelerinde saptanmış olan ilk virüstür. EBV ilişkili gastrik kanser (EBViGK) tüm dünyadaki gastrik kanserlerin yaklaşık %10'nu oluşturur. EBViGK'lerinin genetik temelinde tümör baskılayıcı genlerin promotor bölgesinde DNA hipermetilasyonu sık görülmektedir. EBV enfeksiyonunun metilasyonu nasıl indüklediği tam olarak aydınlatılamamış bir konudur. Bu derlemede, EBViGK'lerdeki gen promotor bölge hipermetilasyonu hakkındaki son literatür gözden geçirilecektir.

Epstein-Barr Virus and DNA Methylation in Gastric Cancer

Öz:
Gastric cancer (GC) is the fourth most common type of malignity and the sec- ond leading cause of cancer death worldwide. Infectious agents such as He- licobacter pylori (H.pylori) and Epstein-Barr virus (EBV) play important role in the etiology in GC. EBV is a double-stranded DNA virus, approximality 184 kb in size, included in Herpes virus family and is the first virus determined in human neoplastic cell. EBV associated gastric cancer (EBVaGC) constitutes approximately 10% of whole GC in wordlwide. DNA hypermethylation in promotor region of tumor supressor genes is seen frequently in the genetic basis of EBCaGC. It is not a clear subject how EBV infection induce the meth- ylation. In this review, the current literature about the gene promotor region hypermethylation in EBVaGCs will be revised.
