miRNA Biyogenezi ve Kanser Patogenezindeki Fonksiyonu

Öz:
Amaç: Küçük kodlamayan RNA molekülleri olan miRNA'lar, yaklaşık 22 nükleotit uzunluğundadırlar. Gen ifadesinin transkripsiyonel ve post-transkripsiyonel düzenlenmesinde fonksiyon gösteren bu moleküller gelişimsel zamanlama, embriyogenez, hücre farklılaşması, organogenez, metabolizma, apoptozis gibi biyolojik süreçlerde ve kanserin de yer aldığı birçok hastalıkta önemli rol oynamaktadır. Kanser gibi proliferatif hastalıkların oluşumunda miRNA disregülasyonunun katkısı olabileceği düşünülmektedir. miRNA'lar hedef genlerinin tümör oluşumundaki rollerine dayanılarak onkogenik ya da tümör baskılayıcı miRNA'lar olarak isimlendirilebilir. Kanserin teşhisine yardımcı olan kan temelli tümör marker'larının özellikle tümör evrelerinin belirlenmesinde düşük duyarlılık gösterdiği bilinmektedir. Düzenleyici mRNA ekspresyon profillerinin aksine, doku miRNA marker'ları kanserin belirlenmesi ve evrelendirilmesinde daha güvenilir olduğu görülmektedir. Bu nedenle dolaşımdaki miRNA'lar kanserin tanı ve prognozu için ideal bir biomarker sınıfı olarak öne çıkmaktadır. ( Sakarya Tıp Dergisi 2016, 6(2):86-93 )

miRNA Biogenesis and Functions on Cancer Pathogenesis

Öz:
Purpose: miRNAs are small non-coding RNA molecules that is approximately 22 nucleotides in length. These molecules which function in transcriptional and post-transcriptional regulation of gene expression play an important role in multiple biological processes, including developmental timings, embryogenesis, cell differentiation, organogenesis, metabolism, apoptosis, and various diseases, including cancers. It is thought that miRNA disregulation leads to development of proliferative disease such as cancer. miRNAs can be termed as oncogenic or tumor suppressor miRNAs based on the role of their target genes on the formation of tumors. Its known that blood-based tumor markers for cancer diagnosis have low sensitivity for identifying tumor stage. Unlike the regulatory mRNA expression profiles, tissue miRNA markers seem to be more reliable in the detection and staging of cancer. Therefore circulating miRNAs represent a class of ideal biomarkers for cancer diagnosis and prognosis. ( Sakarya Med J 2016, 6(2):86-93 )
