Potato Dextrose Agar With Rose-Bengal and Chloramphenicol: A New Culture Medium to Isolate Pathogenic Exophiala dermatitidis From the Environment

Objective: The objective of this study is to describe the potatodextrose agar (PDA) with rose-Bengal and chloramphenicol asa new and simple medium (R-PDA chloramphenicol agar) tofacilitate the detection of black yeast, Exophiala dermatitidiscompared to ready-made conventional medium, namely rose-Bengal chloramphenicol agar.Methods: We prepared a new medium by adding chloramphenicoland rose-Bengal to ready-made PDA.Results: This medium proves better growth of the black yeast interms of increased colony size compared to commercial rose-Bengal chloramphenicol agar. The increase in colony size aidsfor distinguishing the slow growing black yeast from all theother filamentous fungi.Conclusions: Compared to traditional rose-Bengal chloramphenicolagar, R-PDA chloramphenicol agar is superior to isolateExophiala dermatitidis among other fast growing filamentousfungi which are present in the environment samples.Klimik Dergisi 2018; 31(1): 11-5.

Patojen Exophiala dermatitidis’i Çevreden İzole Etmek İçin Yeni Bir Besiyeri: “Rose”- Bengal ve Kloramfenikol Eklenmiş Patates Dekstroz Agarı

Öz:
Amaç: Bu çalışmanın amacı, kullanıma hazır konvansiyonel bir besiyeri olan “rose”-Bengal kloramfenikol agarıyla karşılaştırıldığında, kara maya olarak adlandırılan Exophiala dermatitidis'in saptanmasını kolaylaştıran yeni ve basit bir besiyeri olarak “rose”-Bengal ve kloramfenikol eklenmiş patates dekstroz agarı (PDA)’nı (R-PDA kloramfenikol agarı) tanıtmaktır. Yöntemler: Hazır PDA’ya kloramfenikol ve “rose”-Bengal boyası ekleyerek yeni bir besiyeri hazırlanmıştır. Bulgular: Kara mayanın, bu besiyerinde, ticari “rose”-Bengal kloramfenikol besiyeriyle karşılaştırıldığında, daha büyük koloniler oluşturduğu kanıtlanmıştır. Kolonilerin daha büyük olması, bu yavaş üreyen kara mayanın, diğer ipliksi mantarlardan ayırt edilmesini kolaylaştırmıştır. Sonuçlar: Yeni bir besiyeri olan R-PDA kloramfenikol agarı, yavaş çoğalan Exophiala dermatitidis’in, çevre örneklerinde bulunan diğer hızlı çoğalan ipliksi mantarların arasında izole edilmesi için, geleneksel “rose”-Bengal kloramfenikol agarından daha üstündür.
