GÜMÜŞ NANOPARTİKÜLLERİNİN ÜZÜM ÇEKİRDEĞİ EKSTRAKTININ ENDOTOKSİK KALP DOKUSUNDAKİ HİSTOLOJİK DEĞİŞİKLERE VE TNF-ɑ VE BNP EKSPRESYONUNA ETKİSİ
ipopolisakkarit (LPS) tarafıdan oluşturulan endotokseminin patofiyolojik mekanizmalarının anlaşılmasında ve tedavisinde pek çok strateji geliştiilmesine rağmen, hala yoğun bakım ünitelerinin en önemli problemlerinden biridir. Bu çalışmada, endotoksik şok üzerine üzüm çekirdeği kullanılarak biyolojik olarak sentezlenmiş gümüş nanopartiüllerin (AgNP) koruyucu bir etkisi olup olmadığının kalp dokusundaki histolojik değişiklikler ve beyin natriüreti pepti (BNP) ekspresyonlarındaki değişimler ortaya konularak belirlenmesi amaçlanmıştı. Çalışma için 80 adet Wistar albino cinsi yetikin erkek sıçan 8 eşit gruba ayrılmıştı. Çalışma sonucunda Kontrol ve AgNP uygulanan gruplarda sağlıklı normal bir kalp görüntüsü izlenirken, LPS uygulanan grupta hücre çekirdeklerinde küçülme ve yapısal olarak düzensizlik görülmüştür. LPS ile birlikte AgNP uygulanan gruplarda normale yakın görüntü izlenmişti. Tümör nekroze edici faktör-alfa (TNF-α) ve BNP immünreaktiite sonuçlarının LPS grubunda kontrol grubuna göre istatitisel olarak anlamlı bir şekilde arttı, LPS ile birlikte AgNP uygulanan gruplarda ise artmış olan TNF-α ve BNP immünreaktiitesinin kontrol grubuna benzer şekilde olduğu görülmüştür.
