Tavuk eti örneklerinde genişlemiş spektrumlu beta-laktamaz üreten Escherichia coli

Öz:
Amaç: Bu çalışmada ticari olarak satışa sunulan ambalajlı ve am balajsız tavuk but, göğüs ve kanat etlerinde genişlemiş spektrumlu β-laktamaz (GSBL) üreten Escherichia coli prevalansını tespit etmek amaçlandı. Gereç ve Yöntem: Çalışmada, farklı marketlerde satışa sunulan 30 tavuk but, 30 tavuk göğüs ve 30 tavuk kanat örneği materyal olarak kullanıldı. Her bir numuneden, ön zenginleştirme sonrası Brillance ESBL agara ekimler yapılmış, uygun inkübasyonun ardından üre yen şüpheli kolonilerden max. 5 tanesi seçilerek, EMB agara geçildi. İzolatların E.coli olarak identifikasyonunda IMViC testler kullanıldı. Suşların GSBL üretim özellikleri Klinik ve Laboratuvar Standartları Enstitüsü (CLSI)’nce belirtilen kombine disk difüzyon yöntemi ile fenotopik olarak belirlendi. Bulgular: Tavuk eti örneklerinden elde edilen 159 E. coli şüpheli izolattan, 152 tanesi (%95,5) E.coli olarak belirlendi. Kombine disk difüzyon testi sonrasında 28 (%18,4) izolatın GSBL pozitif olduğu tespit edildi. İncelenen göğüs etlerinde 17 (%60), but örneklerinde 9 (%32) ve kanat örneklerinde de 2 (%7) adet GSBL izolatı bulundu. GSBL üreten E.coli izolatlarının ambalajlı ve açık örneklerdeki dağılı mı, sırasıyla %71 ve %28 olarak belirlendi. Öneri: Tavuk etlerinde GSBL üreten E. coli varlığı, bu tip direncin insanlara aktarımında gıdaların da önemli bir kaynak olabileceğini ortaya koymaktadır. Gıdalar düzenli olarak patojen bakteriler kapsa mında incelenmesine rağmen, GSBL üreten bakteriler bu kapsamda değildir. Bu durum gıda zinciri vasıtasıyla yeni dirençli bakterilerin fark edilmeden artmasına neden olabilmektedir. Bu nedenle bakteri lerde fenotipik olarak direncin belirlenmesinin yanı sıra, elde edilen izolatların ileri analizlerle genotipik olarak da bu özelliği taşıyıp ta şımadığının tespit edilmesi önerilmektedir.

Öz:
Aim: The aim of this study was to determine the prevalence of ex tended spectrum β-lactamase (ESBL) producing E.coli in retailed chicken thigh, breast and wing meats, with and without packaging. Materials and Methods: In the study, retailed 30 chicken thigh, 30 chicken breast and 30 chicken wing samples, were used as materials. After pre-enrichment, each sample was plated to the Brillance ESBL medium and incubated. Up to 5 suspected colonies were selected and plated to the EMB medium. Then the isolates were identified as E. coli by IMViC test. In order to determine the ESBL production of the isolates phenotypically combined disc diffusion test was perfor med according to the CSLI. Results: From the 159 suspicious isolates, 152 (95.5%) of them identified as E.coli. According to combined disc diffusion test 28 (18.4%) of the isolates were ESBL positive. 17 (60%) ESBL produ cing isolates were found in the breast meat samples while 9 (32%) isolates from thigh and 2 (7%) isolates from wings were positive for ESBL production. The distribution of ESBL producing E. coli isolatesin packaged and unpackaged samples were determined as 71% and 28%, respectively. Conclusion: The presence of ESBL producing E. coli in chicken meats indicates that foods may also be an important source for the trans mission of this resistance to human. Although foods are regularly examined in the context of pathogenic bacteria, bacteria producing GSBL are not covered by this.
