KOAGÜLAZ-NEGATİF STAPHYLOCOCCUS (KNS) VE MACROCOCCUS CASEOLYTICUS SUŞLARININ BİYOFİLM OLUŞTURMA ÖZELLİKLERİNİN VE ADEZYON FAKTÖRLERİNİN ARAŞTIRILMASI

Öz:
Bu çalışmada geleneksel fermente Türk sucuklarından izole edilmiş toplam 50 adet koagulaz-negatifStaphylococcus (KNS) ve 10 adet Macrococcus caseolyticus suşlarının biyofilm oluşturma özellikleri fenotipik olarakbelirlenmiş ve aynı zamanda suşlarda biyofilm oluşumu ile ilişkili genler ve adezyon genlerinin varlığımultipleks polimeraz zincir reaksiyonu (mPZR) ile araştırılmıştır. Fenotipik olarak slaym (slime) üretimiCongo Red agar (CRA), biyofilm üretim düzeyi mikrotitre plaka yöntemi ile belirlenmiştir. CRA besiyerindeslaym üreticisi olduğu belirlenen 33 suşun tamamının mikrotitre plaka yöntemi ile biyofilm üreticisi olduğutespit edilmiştir. KNS ve M. caseolyticus suşlarının 28’inin orta, 23’ünün zayıf ve 1’nin güçlü biyofilm üreticisiolduğu saptanmıştır. Sekiz suşta ise biyofilm üretiminin olmadığı tespit edilmiştir. mPZR denemeleri sonucubiyofilm üreticisi olduğu belirlenen KNS ve M. caseolyticus suşlarının %84.62’sinin araştırılan genlerden en azbirini içerdiği bulunmuştur. Suşların %66.67’sinde eno geni, %15.67’sinde icaA geni varlığı belirlenmiştir.Diğer taraftan biyofilm üreticisi olduğu belirlenen 8 suşun ise çalışma kapsamında araştırılan biyofilmgenlerinin hiçbirini içermediği tespit edilmiştir.

Öz:
In this study, biofilm forming properties of 50 coagulase-negative Staphylococcus (CNS) and 10 Macrococcus caseolyticus strains isolated from traditional fermented Turkish sausages (sucuk) werephenotypically determined and also the presence of genes associated with biofilm formation and adhesion genes in the strains was investigated by multiplex polymerase chain reaction (mPZR). Slime production was determined on Congo Red agar (CRA) by phenotypically and biofilm production level was determined by microtiter plate method. All 33 strains determined to be slime producers in CRA medium were determined to be biofilm producers by microtiter plate method. It has been determined that 28 of KNS and M. caseolyticus strains are medium, 23 are weak and 1 is strong biofilm producers. Biofilm production was not detected in eight strains. As a result of mPZR experiments, 84.62 % of CNS and M. caseolyticus strains determined to be biofilm producers were found to be contain at least one of the genes investigated. eno gene was detected in 66.67% of the strains and the presence of icaA gene in 15.67% of the strains. On the other hand, it was determined that 8 strains determined to be biofilm producers do not contain any of the biofilm genes investigated within the scope of the study.
