Vitamin D uygulamasının endometrium üzerine etkisi

Öz:
Amaç: Vitamin D, mineral metabolizması, hücre çoğalması-farklılanması, apoptoz, immün sistemde görev alanbir hormondur. Endometrium Vitamin D için hedef organlardan biridir. Çalışmamızda Vitamin D uygulamasınınendometrium apoptozisi üzerine etkisini göstermeyi amaçladık.Gereç ve yöntem: Onsekiz sıçan rastgele olarak 3 gruba ayrıldı: Kontrol (grup 1, n:6), 0,5 μg/kg Vitamin D (grup2, n:6), 1 μg/kg Vitamin D (grup 3, n:6) olarak belirlendi. Vitamin D haftada 3 gün, 8 hafta intraperitoneal olarakverildi. Deney bitiminde uterus örnekleri alındı. Apoptozun değerlendirilmesi için Terminal deoxynucleotidyltransferase dUTP nick end labelling (TUNEL), Bax, Bcl-2 ve Kaspaz-3 immunohistokimyasal olarak incelendi.Bulgular: Lümen ve bez epitelinde TUNEL pozitif hücre sayısı grup 1’de grup 2 ve grup 3’e göre anlamlı olarakyüksekti. Bax ekspresyonu lümen epiteli, bez epiteli ve stromal hücrelerde farklılık göstermedi. Lümen ve bezepitelinde Bcl-2 ekspresyonunun Vitamin D uygulanan gruplarda fazla olduğu görüldü. Bununla birlikte stromalhücrelerde grup 3’te Bcl-2 ekspresyonu grup 2 ve grup 1’e göre anlamlı olarak fazla olduğu saptandı. Kaspaz3 ekspresyonu grup 1’in lümen ve bez epitelinde diğer iki gruba göre daha yüksek bulundu. Stromal hücrelerdegrup 1 ve grup 2’deki ekspresyonun grup 3’e nazaran daha fazla olduğu belirlendi.Sonuç: 0,5 μg/kg Vitamin D ve 1 μg/kg Vitamin D uygulaması endometriumda lümen ve bez epitelinde apoptozisigeriletmiştir. Özellikle 1 μg/kg Vitamin D dozunun tüm endometriumda etkili olduğu görülmüştür

Effect of vitamin D application on the endometrium

Öz:
Purpose: Vitamin D is a hormone involved in mineral metabolism, cell proliferation-differentiation, apoptosis, and immune system. Endometrium is one of the target tissue for Vitamin D. We aimed to determine the effects of Vitamin D on apoptosis in the endometrium. Materials and methods: Eighteen rats were randomly divided into three groups, as follows:control group (group 1, n=6), 0,5 μg/kg Vitamin D -treated group (group 2, n=6), and 1,0 μg/kg Vitamin D -treated group (group 3, n=6). Vitamin D was given intraperitoneally three times a week for eight weeks. Uterus samples were obtained from the rats. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL), Bax, Bcl-2, and Caspase-3 were used immunohistochemically for the evaluation of apoptosis. Results: The number of TUNEL positive cells in the luminal and glandular epithelium was significantly higher in group 1 than in groups 2 and 3. Bax expression didn’t differ in luminal epithelium, glandular epithelium, and stromal cells. Bcl-2 expression in luminal and glandular epithelium was higher in groups treated with Vitamin D. However, Bcl-2 expression in stromal cells was significantly higher in group 3 than in group 2, and 1.Caspase 3 expression was higher in the luminal and glandular epithelium of group 1 than the other groups. Caspase 3 expression in group 1 and group 2 was higher in stromal cells compared to group 3. Conclusion: Treatment with 0.5 μg/kg and 1 μg/kg Vitamin D regressed apoptosis in the luminal and glandular epithelium. In particular, the dose of 1 μg/kg was effective on the entire endometrium.
