Akut İshalli Neonatal Buzağılarda Plazma Sitrulin Konsantrasyonunun Değerlendirilmesi

Öz:
Akut İshalli Neonatal Buzağılarda Plazma Sitrulin Konsantrasyonunun Değerlendirilmesi Bu çalışmada sağlıklı ve akut ishalli neonatal buzağılarda plazma sitrulin konsantrasyonu ve ilişkili parametrelerin değerlendirilmesi amaçlandı. Çalışmada 10 sağlıklı, 60 akut ishalli olmak üzere her iki cinsiyetten toplam 70 neonatal (<28 günlük yaş) Holstein ırkı buzağı kullanıldı. Klinik ve laboratuvar bulgular temelinde ishalli buzağılar etiyolojiye göre enfeksiyöz (n=50) ve non-enfeksiyöz (n=10), ishalin şiddetine göre hafif (n=15), orta (n=25) ve şiddetli (n=20), sistemik yangısal yanıt sendromu (SIRS) kriterlerine göre SIRS pozitif (n=27) ve SIRS negatif (n=33) olarak alt gruplarda değerlendirildi. Sağlıklı buzağılarda neonatal dönem plazma sitrulin konsantrasyonu ve ilişkili parametrelerde yaşa bağlı değişikliklerin belirlenmesi amacıyla örneklemeler 2., 7., 14., 21. ve 28. yaĢam günlerinde, shalli buzağılarda ise bir kez yapıldı. Dışkı örneklerinde etiyolojik değerlendirme; Cryptosporidium parvum, Rotavirus, Coronavirus, E. coli K99 ve Giardia duodenalis antijenlerine yönelik hızlı test kiti, parazitolojik ve mikrobiyolojik muayene ile gerçekleştirildi. ishal ile ilişkili rutin hemato-biyokimyasal analizler, tam kan sayımı (WBC, RBC, HCT, PLT), kan gazları (pH, PCO2, PO2, Na, K, HCO3, BE) ve serum biyokimyasal parametreleri (AST, GGT, üre, kreatinin, total protein, albumin) kapsadı. Plazma sitrulin konsantrasyonu yanında, sitrulin ile yakın iliĢkili arjinin, nitrik oksit ve amonyak konsantrasyonları ölçüldü. Oksidatif durumu değerlendirme amacıyla da Total Oksidan Seviye (TOS) ve Total Antioksidan Kapasite (TAK) analizleri gerçekleştirildi. Sağlıklı buzağılar göre, akut ishalli buzağılarda plazma sitrulin ve arjinin konsantrasyonları ile TAK değerinin istatistiksel anlamlı düşük, kan amonyak ve plazma nitrik oksit konsantrasyonu ile TOS değerinin ise yüksek olduğu saptandı. Plazma sitrulin konsantrasyonundaki azalmaların etiyolojiye göre enfeksiyöz, ishalin şiddetine göre hafif, orta ve şiddetli, SIRS değerlendirmesine göre ise SIRS pozitif gruplarda olduğu belirlendi. Plazma arjinin konsantrasyonunun enfeksiyöz, şiddetli ve SIRS pozitif gruplarda azaldığı görüldü. Nitrik oksit düzeyi enfeksiyöz, hafif, orta, şiddetli ve SIRS pozitif gruplarda yüksek bulundu. Amonyak düzeyindeki artışların enfeksiyöz, şiddetli ve SIRS pozitif gruplarda olduğu ortaya konuldu. TOS seviyeleri akut ishalli tüm gruplarda yüksek, TAK seviyeleri ise düşük bulundu. Sonuç olarak, akut ishalli buzağılarda plazma sitrulin ve arjinin konsantrasyonlarında azalma ile kan amonyak düzeyinde artış olduğu ilk defa belirlendi. Sitrulin konsantrasyonunun özellikle enfeksiyöz grupta azalması ile akut ishalli buzağılarda sitrulinin bağırsak hasarını dolaylı olarak yansıtabileceği gösterildi. Akut ishalli neonatal buzağılarda tedavide sitrulin uygulamasının endike olabileceği ve sonuçların gerçekleştirilecek çalışmalara temel oluşturabileceği kanısına varıldı.

Akut İshalli Neonatal Buzağılarda Plazma Sitrulin Konsantrasyonunun Değerlendirilmesi

Öz:
Evaluation of Plasma Citrulline Concentration in Neonatal Calves with Acute Diarrhea In this study, it was aimed to evaluate plasma citrulline concentration and related parameters in healthy and neonatal calves with acute diarrhea. A total of 70 neonatal (<28 days of age) Holstein calves of both sexes, comprising 60 with acute diarrhea and 10 healthy were enrolled in the study. On the basis of clinical and laboratory findings, calves with diarrhea was classified as infectious (n=50) and non-infectious (n=10) according to etiology, mild (n=15), moderate (n=25) and severe (n=20) according to severity, SIRS positive n=27) and negative (n=33) according to the criteria of the systemic inflammatory response syndrome (SIRS). Etiological evaluation was provided with parasitological, microbiological examination and rapid test kit analysis against antigens of Cryptosporidium parvum, Rotavirus, Coronavirus, E. coli K99 and Giardia duodenalis in stool samples. Routine haemato-biochemical laboratory analysis related with diarrhea was covered by complete blood count (WBC, RBC, HCT, PLT), blood gases (pH, PCO2, PO2, Na, K, HCO3, BE) and serum biochemical parameters (AST, GGT, urea, creatinine, total protein, albumin) measurement. Besides the concentration of plasma citrulline, arginine, nitric oxide and ammonia concentrations that closely related with citrulline were determined. Total Oxidant Status (TOS), Total Antioxidant Capacity (TAC) analysis were performed in order to evaluate the antioxidant properties of citrulline. Plasma citrulline and arginine concentrations and TAC levels were found to be lower while concentrations of blood ammonia and plasma nitric oxide concentrations and TOS levels were higher in calves with acute diarrhea compared to healthy calves. The reductions in plasma citrulline concentrations were found to be in infectious, mild, moderate and severe diarrhea, and SIRS positive groups. Plasma arginine concentration decreased in infectious, severe and SIRS positive groups. Nitric oxide levels were high in infectious, mild, moderate, severe and SIRS positive groups. Increases in ammonia levels were found to be in infectious, severe and SIRS positive groups. TOS levels were high and TAC levels were low in all groups with acute diarrhea. As a result, the decreased concentrations of plasma citrulline and arginine, and the increase in blood ammonia level were firstly determined in calves with acute diarrhea. It has been shown that plasma citrulline concentration, especially in the infectious group, may indirectly reflect intestinal damage in acute diarrhea. It’s concluded that the effect of citrulline administration might be documented in neonatal calves with diarrhea based on these results.
