Su kabağı (Lagenaria siceraria) Genetik Kaynaklarının (Çekirdek Koleksiyonunun) Tuz Stresine Karşı Taranması (Screening)

Öz:
Bu çalışmada, ülkemiz su kabağı genetik kaynaklarından seçilmiş olan 150 su kabağı genotipi 10 dS/m tuz stresine karşı testlenmiştir. Toplam taze ağılık bakımından karpuz çeşitlerinden ve ticari su kabağı anaçlarından daha iyi sonuç veren genotip sayısı sırasıyla 121 ve 57olurken, 3 genotip Ferro anacından daha iyi sonuç vermiştir. Toplam kuru ağırlık bakımından ise 99 genotip karpuz çeşitlerinden, 121 genotip su kabağı anaçlarından ve bir genotip Ferro’dan daha iyi sonuç verirken, taze ve kuru ağırlık açısından RS841’den daha iyi sonuç veren genotip bulunmamıştır. Tolerant (5) ve duyarlı (5) olarak seçilen genotiplerde yapılan çalışmada, tuz stresine bağlı olarak bitki organlarındaki Na + ve Cl - içeriklerinin arttığı ve artışın kökte ve gövdede daha fazla olduğu görülmüştür. Gövdenin Na + ve Cl - içerikleri yaprak ve köklerden daha yüksek bulunmuştur. Tuz stresi altında yaprak oransal su içeriği, oransal iyon sızıntısı ve malondialdehit miktarı ile bitkisel gelişim arasında negatif bir ilişki olduğu görülmüştür. Stres koşulları altında prolin, gilisnbetain ve enzim aktivitelerinde önemli artışlar görülmüştür. Su kültüründe yapılan çalışmada tuz stresinden kaynaklı biyokütledeki azalma aşılı bitkilerde daha az olmuştur. Aşılı bitkilerin yaprak alanı (%4-83), taze (%4-99) ve kuru (54-169) ağırlıkları aşısızlardan daha yüksek olmuştur. Yaprak alanı ve yeşil aksam taze/kuru ağırlığı bakımından en yüksek değerler 70-07 üzerine aşılı bitkilerden alınmıştır. Bitkilerin köklerindeki Na + konsantrasyonu yapraklardan daha yüksek bulunmuş, yüksek biyokütleye oluşturan aşı kombinasyonlarının Na + içeriği daha düşük bulunmuştur. Tuz stresi koşulları altında yaprakların prolin ve glisinbetain içerikleri ile SOD, CAT, APX ve GR aktivitelerinde anaçlara bağlı önemli artışlar tespit edilmiş fakat tolerans seviyesi ile anlamlı bir ilişki bulunmamıştır. Aşılı bitkilerde erkek ve dişi çiçekler sırası ile 5 ile 4-8 gün daha erken açmışlardır. Tuz stresi ile birlikte SPAD ve fotosentez miktarının azaldığı ve bu azalmanın stresin ilerleyen zamanlarında daha belirgin olduğu görülmüştür. Tuz stresi altında, C.T./42-09, C.T./45-07 ve C.T./47-02 aşı kombinasyonları dışındakiler %10 ile %92 daha fazla gövde+yaprak taze ağırlığı ve yerel genotiplerden 35-01, 42-05, 56-01, 70-04 ve 70-07 üzerine aşılı olan bitkiler kontrol bitkilerinden daha yüksek (%22-88) gövde+yaprak kuru ağırlığına sahip olmuştur. Tuz stresi ile birlikte meyve ağırlığı azalmış ve en küçük meyveler aşısız kontrol bitkilerinden hasat dilmiştir. Tuz stresi altında en ince kabuklu meyveler kontrol bitkilerinden alınmıştır. Meyve eti sertliği 8.3-12 N arasında değişmiştir. SÇKM anaçlara bağlı olarak değişmiş (11.1-12.3) ve bütün uygulamalar karpuz için tüketilebilir sınırlar içinde SÇKM değerlerine sahip olmuşlardır. En düşük skala değeri (7.2) C.T./RS841 kombinasyonunda tespit edilirken, diğer aşı uygulamaları 7.9 ile 8.3 arasında değerler almışlardır. Bütün aşılı uygulamaları kontrol koşuları ve tuz stresi altında kontrol bitkilerinden daha yüksek verim değerlerine sahip olmuşlardır.
